The present work shows computationally and experimentally that ENOblock acts synergistically with colistin, a last-resort antibiotic for A. baumannii. This finding is significant as combination therapies can help overcome resistance and enhance the effectiveness of existing drugs.

BCP reveals that ENOblock induces unique morphological changes in A. baumannii compared to conventional antibiotics targeting major cellular pathways. While it appears to disrupt membrane integrity, its profile is different from the membrane-targeting antibiotic colistin.

ENOblock significantly reduced A. baumannii adherence/invasion in epithelial and macrophage cells probably through the inhibition of enolase binding to host proteins, and improved survival rates in G. mellonella infected with A. baumannii, demonstrating its therapeutic potential.

Computational docking indicated strong binding of ENOblock to enolase, which was confirmed by ITC analyses of wt & enolase mutants, and assays with Δeno strain. The Δeno strain exhibited a 4-fold increase in MIC and greater growth resistance to ENOblock, supporting enolase as its probable target.

SYTOX Green assay revealed that ENOblock caused rapid and extensive membrane damage within 10 minutes of exposure. Moreover, BCP analysis revealed unique morphological changes not observed with clinically used antibiotics, suggesting a distinct ENOblock MoA.

Key insights: ENOblock demonstrated potent antibacterial activity with an MIC50 of 16 mg/L against clinical carbapenem- and colistin-resistant A. baumannii strains, surpassing the MIC50 values of colistin and imipenem/meropenem.
