a couple billion years ago O2 levels started rising and oxidizing everything within biological systems. intracellular redox pathways evolved to maintain redox states inside the cell but outside was still perilous. this is why there are very few -SH cysteines on surface proteins.

i spend a lot of time discussing mild traumatic brain injury (mTBI) with my colleagues due to the nature of my work so i think it's funny that i get a lot of Myers-Briggs Type Indicator (MTBI) advertisements. myers briggs is complete nonsense btw.

in grad school we rescued a black widow from the elevator. we named her princess and kept her in an erlenmeyer with glass pipettes for years. we fed her crickets every week and sometimes the odd mosquito.

if you want to understand how natural products became medicines you have to understand the coevolutionary history of the secondary metabolites and the proteins that they target. this history is impossibly rich.

by identifying covariation hotspots in GPCRs we can functionally couple residues that bind with the receptor ligand and intracellular functional loops (like g-protein or beta arrestin binding domains).

with evolution-guided pharmacology we can non-conserved contacts between gpcrs and their peptide hormone ligands, and identify functional covariation hotspots on the intracellular side of the membrane. this guides us to active site residues responsible for information transmission

it’s been almost 20 years since i set up my first chemical reaction. it's still magical to me that by understanding chemistry you can fundamentally change physical matter. rearrange the same atoms in the right way, and you change the practical and monetary value of a molecule.

choosing a drug target is the most important decision of a drug discovery campaign. of 704 proteins targeted by FDA-approved drugs, 95% predate mammals evolutionarily, 35% predate animals (eukaryotes or older) 5% are mammal specific or younger, and the mediate age of the gene is 680 million years

i think i figured out how to detoxify poison oak. the poison in poison oak (urishiol) gets absorbed by the skin and the quinone of urishiol is electrophilic and covalently modifies proteins. the modified peptide gets displayed on MHC and causes an immune response. that's how the poison works.
