To ensure that propagation results weren’t reflecting previously-reported effects of psychedelics, we evaluated propagations alongside functional connectivity and activity autocorrelation. Brain and self-report associations with propagations were dissociable.

Psychedelics also decreased the magnitude of cortical propagations (irrespective of hierarchical directionality) broadly across both imaging modalities/species.

To establish generalizability across a broad definition of psychedelics, we also evaluated psilocybin and LSD. To stress-test these findings, we evaluated 9 case-control comparisons across studies and imaging modalities. Effects were largely consistent.

To our surprise, MDMA reduced bottom-up propagations into the DMN.

Then we used ∇DMN as a set of reference directions to contextualize whether movement was into (blue, “bottom-up”) or out from (red, “top-down”) the DMN.

Specifically, we tested the idea that psychedelics increase bottom-up processing in the default mode network (DMN). In humans and mice, we used optical flow to describe the movement of cortical activity as vectors.
