‘Microbiome-Based Therapeutics for Insomnia’
Figure 1, the basis for microbiome-based therapeutics for insomnia lies in the relationship between gut microbiota and the brain. The gut microbiota influences sleep through the brain–gut microbiota axis, a bidirectional communication system that consists of the hypothalamic–pituitary–adrenal axis, microbiota metabolites, the immune system, and the vagus nerve.

Parkinson’s ‘[Curcumin Nanoparticle] CNP treatment, both pre and post PQ exposure, may exert a protective (or more accurately, restorative) effect on mitochondrial morphology, especially on the cristae, which are important for ATP production’ Mitochondrial Morphology:
‘As shown in Fig. 5A, in the untreated SH-SY5Y cells, mitochondria appeared to be elongated in shape, had a well-defined double membrane with the inner membrane exhibiting cristae, all of which are characteristics of healthy mitochondria.
Treatment with [Paraquat] PQ (Fig 5B), however, demonstrated swelling of mitochondria, and loss of and/or damage to the cristae.
Treatment with [Curcumin loaded Nanoparticles] CNPs (Fig 5C) did not appear to change mitochondrial morphology compared to the untreated cells (Fig 5A), although slight changes in the cristae do appear as shown by the spaces in the intermembrane space.
CNP post-treatment group (Fig 5D) presented loss of the cristae but not as severe as treatment with PQ only (Fig 5B).
Similarly, CNP pre-treatment group (Fig 5E) also indicated loss of the cristae, evidently more than CNP post-treatment (Fig 5D), with some mitochondria displaying a rounded shape, although not as severe as treatment with PQ only.

This indicates that CNP treatment, both pre and post PQ exposure, may exert a protective (or more accurately, restorative) effect on mitochondrial morphology, especially on the cristae, which are important for ATP production.’

Cohort columns indicate the number of subjects (n) and prevalence (%) of individuals who are positive for dUTPase Abs against a single or multiple herpesviruses or negative. A positive reaction was defined as a serum sample that led to a signal twice over the background OD of the control serum (95% Confidence interval). An individual was considered positive for antibodies to a specific dUTPase if a positive reaction occurred in 25% of the serum/plasma samples’

‘SARS-CoV-2, in particular, has had an overwhelming global impact [91, 92]. MCs and basophils play critical roles in COVID-19 pathogenesis. Basophils drive the antibody response, while MCs form the first line of defense against SARS-CoV-2, contributing to a hyperinflammatory environment and triggering the cytokine storm. Through histamine release, MCs overexpress IL-4, IL-6, IL-8, and other proinflammatory cytokines, leading to lung hyperinflammation, microvascular leakage, matrix degradation, angiogenesis, and pro-coagulation [93]. Alterations in the host microbiota appear to significantly impact “long-COVID” or “Post-COVID syndrome” (PCS). PCS often progresses in close association with mast cell activation syndrome (MCAS). Specifically, SARS-CoV-2 infection can trigger autoimmune reactions that activate MCs, causing hyperinflammation and MCAS, which, in turn, mediate PCS symptoms [94, 95]. Given these pathophysiological considerations and the potential complexity of interactions between microbiota and dysfunctional MCs in MC activation disease, altered microbiota-MC crosstalk during SARS-CoV-2 infection may contribute to the PCS-MCAS link’

‘To understand if chronic hyperactivation of DA neurons is sufficient to cause neurodegeneration, we developed a chemogenetic mouse model. Our results indicate that chronically increasing neural activity in midbrain DA neurons results in alteration of circadian locomotion patterns, and prolonged activation leads to selective degeneration of SNc axons and eventual death of midbrain DA neurons. These changes were accompanied by altered intracellular calcium dynamics and transcriptomic changes consistent with calcium dysregulation, supporting a role for increased neural activity in driving neurodegeneration in PD.’

‘microbiota‐gut‐brain axis.. We hypothesized that the restricted type & amount of food intake characteristic of ARFID diminishes gut microbial diversity, including beneficial bacteria & their metabolites capable of signaling to the brain, to modulate biopsychological pathways relevant to ARFID..’
‘The pathophysiological microbiota‐gut‐brain‐axis model of avoidant/restrictive food intake disorder (ARFID). Evidence suggests that the microbiota‐gut‐brain‐axis plays a pathophysiological role in the onset and maintenance of symptoms of ARFID. A restricted diet in amount and variety of foods, including fiber and protein‐rich foods, promotes a specialized microbiota, thus depleting microbial diversity and their metabolites, including short‐chain fatty acids (SCFAs) and vitamins. High amounts of refined/processed sugar increase peripheral inflammation. Bidirectional biopsychological pathways through which these nutrition‐induced effects on the gut microbiota contribute to ARFID phenotypes include: homeostatic intake, food reward, interoception, sensory sensitivity, disgust, perseveration, fear‐based learning, and mood. Candidate biological mechanisms include vagal, inflammatory, and endocrinological signaling via microbial metabolites. GLP‐1: glucagon‐like peptide 1. HPA: hypothalamic–pituitary–adrenal. PYY: peptide YY.’

‘Figure 1. The basic model, highlighting the potential for feedback loops that further reinforce the initial insult. Mechancial deformation of the brainstem could be triggered by heredetary connective tissue disorders, physical trauma, or acute infection (mediated by connective tissue degradation as part of the normal immune response). This deformation alters brainstem signaling in key regions such as the cNST, which triggers a state of chronic hyperinflammation, leading to further expression of connective-tissue degrading factors, mast cell activation disorders, and latent viral reactivation, which further reinforce the loop.’

PEM not specifically noted but the effects of PEM possibly referenced here? (Mice) 🤔
‘Interestingly, sex-specific analysis revealed that infection more adversely affected female than male behaviour. This impairment was partially mitigated by WEHI-P8 treatment, whereas PLT treatment showed no benefit (Supplementary Fig. 9k). To further examine behavioural differences between males and females, we analysed the recognition index, defined as the proportion of time spent exploring the novel object relative to the total exploration time. This metric did not reveal significant sex- or drug dependent differences upon infection (Fig. 5h). However, the overall distance travelled during behavioural tests was significantly reduced in infected females, but not males. Notably, this effect was rescued by WEHI-P8 treatment but not by PLT (Fig. 5i). Overall, our findings suggest that female animals are more susceptible to PASC-related cognitive symptoms, which appears to be a further similarity to humans56.‘
