SPROUTS_DB was developed via proteomics analysis of complete medium with EV-depleted FBS – but without cells – mimicking all the passages for EV purification. SPROUTS_DB includes 1288 entries, representing soluble and EV bovine proteins, plus common laboratory contaminants. (4/5)

Many peptides are evolutionarily conserved and this strategy would non-specifically remove both contaminants and proteins of interest. SPROUTS_DB is a tailored database when FBS is the noisy contributor in the game, lowering the risk of losing "good" candidate proteins. (3/5)

In vitro models frequently rely on fetal bovine serum (FBS) for cell culture, introducing bovine-derived protein contaminants, including residual FBS-EVs, which can lead to false-positive identifications, especially among low-abundance proteins. Why not simply filter out bovine peptides? (2/5)
