Does every enhancer work with every promoter? With @jengreitz.bsky.social and Will Greenleaf, we revisit this long-debated question and resolve an outstanding contradiction in the field. A tour 🧵👇 www.biorxiv.org/content/10.6...

Intrinsic promoter responsiveness dictates sensitivity to transcriptional activation by enhancers
Enhancers activate specific target promoters, but whether intrinsic enhancer-promoter compatibility contributes to this specificity is debated. Recent studies using different reporter assays have reac...

Great experience compiling years of collective screening experience to produce what we hope is a comprehensive protocol for others to accessibly scale up experiments! And some fun new data hidden in there :) @joshtycko.com www.nature.com/articles/s41...

We wrote a perspective "How to build the regulatory genome: a constructionist guide to the cis-regulatory code", out in Development yesterday. Title says it all. Find it here: journals.biologists.com/dev/article/...

Thanks to co-authors Charles C. Bell, Brendan R. Camellato, M. Joaquina Delás, Daniel M. Ibrahim, and @michaeltmont.bsky.social And also to all the participants of the 2024🤨 Company of Biologists meeting that inspired this.
Participants of the meeting posing on a staircase looking fantastic.

Had a lot of fun writing this “tools of the trade” highlight for our Variant-EFFECTS technology. Check it out! 🛠️

Very cool work! I'm particularly excited about the integration of multiple models/prediction heads in this approach, given how many aspects of gene regulation likely need to be considered to achieve a specific quantitative design goal.

I am elated to share that our manuscript describing Variant-EFFECTS, a high-throughput technology we developed to precisely and quantitatively measure the effects of CRISPR-mediated edits on gene expression, is now published at @cellpress.bsky.social: authors.elsevier.com/c/1kxgiL7PXu...
