Intriguingly, pharmacological PRC2 inhibition in human cortical organoids induces increased production of the ECM components Syndecan 1 and Laminin alpha 1. (9/11)

Our cell type-specific profiling of the genome-wide H3K27me3 profile not only identified neuronal differentiation, but also extra-cellular matrix (ECM) genes as targets of PRC2. (7/11)

We found that loss of H3K27me3 in human cortical organoids induces a shift of neural progenitor cell proliferation towards differentiation that results in reduced organoid size. (5/11)

Here, we performed pharmacological inhibition of PRC2 in human cortical organoids from week 3 to 8, leading to a near complete loss of H3K27me3, and studied NPC behaviour. (4/11)

We apply an innovative, highly multiplexed approach, Epi-CyTOF, to study > 30 epigenetic modifications at single-cell level in the human developing neocortex. We identify H3K27me3, deposited by PRC2, as the modification with the strongest cell type-specific enrichment. (2/11)

How do epigenetic modifications contribute to human #neocortex #development? Excited to see our work out in #Neuron. We applied Epi-CyTOF and identified a novel role for PRC2 in regulating #ECM. @crtd-tud.bsky.social @cp-neuron.bsky.social www.sciencedirect.com/science/arti... A thread below 👇

Metabolic and epigenetic regulation are tightly linked. The deposition and removal of epigenetic marks, including DNA and histone modifications, requires substrates provided by a plethora of metabolic pathways.

Mutations in various histone modifiers cause developmental disorders linked to impaired brain development. These disorders highlight the importance of activating and repressive histone modifications in controlling human cortical development.

The neocortexes of mice and humans differ in various respects, including developmental timing. Epigenetic regulation has emerged as an important mechanism to control developmental timing.
