This paper is not evidence that LC and ME/CFS have a shared cause; no diagnostic biomarker has been validated, and no therapeutic target has been identified. It only suggests both diseases occupy overlapping regions of immune-metabolic space in this computational framework. 6/6

Pathway enrichment and network biology studies in general have known limitations. Different gene sets can map onto the same large immune/metabolic network even when the underlying biology is not truly shared. 5/6

The idea that chronic inflammatory diseases converge on immune/metabolic stress pathways is biologically plausible. Common hubs are present in many chronic diseases. Finding them does not necessarily reveal a specific disease mechanism. 4/6

A lot of convergence on immune/cytokine signalling, mitochondrial biology, metabolism and neuroendocrine signalling is claimed, but there is limited overlap in the actual genes identified in each disease. The authors do not claim that the diseases are identical. 3/6

Digging a bit deeper; the single patient reporting initial improvement (which had to be stopped due to side effecct and compensatory mechanisms kicking in) was also an author on the study. GWAS studies with ME/CFS or LC did not identify rs5522. If it was that important, it would have been.

Much is worth exploring. But there is very little evidence for this. A case report of one homozygous patient. There is no reported genetic link with this receptor, there are no clinical trials or other strong data It's potential importance is hugely overstated in light of this.

If this is plague, it can be treated very well with antibiotics. There are regular outbreaks annually (~2000 deaths), and that is not an pandemic risk. Unfortunately there is no information on antibiotic use. The sad death of at least one person should have resulted in
