It’s preprint week in the Koutmou lab! With Lyle Simmons’ lab, we identify rRNA methyltransferases in a Gram-positive bacterium, assign functions to 11 uncharacterized B. subtilis genes, and show how chemical modifications shape ribosome assembly and antibiotic sensitivity. doi.org/10.64898/202...

New preprint from Kaley Simcox in the Koutmou lab! She developed a HILIC-MS/MS approach for total tRNA sequencing, achieving 75–100% coverage for most isoacceptors - without ion-pairing reagents. This enabled the discovery of new modified positions on E. coli tRNAs. doi.org/10.64898/202...

New preprint from the Koutmou, Jain & Wiedenheft labs! We identify sub-stoichiometric pseudoU sites in MS2 RNA genome and benchmark Nanopore DRS with LC-MS/MS + CLAP. Takeaway: broad mod trends can inform biology, but individual calls/occupancy need orthogonal validation. doi.org/10.64898/202...
