I think the main issue that's happened with this comic is that the person posting it on Twitter seems to have been a chaser so wasn't trustworthy, people on twitter didn't know Poke was trans, and finally they just don't understand Spidey outside of the films where their relationship with 1/2

I mean people absolutely do invalidate the suffering that trans people go through, not about needing to hate yourself it's just something that happens in different ways for different people. I feel like saying their own biases isn't the right way of wording it, definitely misinterpreting it but from what I've seen it's mostly other trans woman so more likely it's their fears and over cautiousness because of...well *vaguely gestures at the world*
