Couldn’t be prouder of my big sister! Her paper on proPE — a Prime Editing system with prolonged editing window — just came out in Nature Catalysis! By using two RNAs instead of one, proPE expands prime editing to previously unreachable, disease-linked SNPs. www.nature.com/articles/s41...

Prime editing is a CRISPR methodology whose efficiency declines with distance from the target sequence. Here the authors demonstrate prime editing with prolonged editing window, proPE, which extends the editing distance, enabling the use of prime editing for therapeutic interventions.

Single-cell electroporation is performed via patch pipette and under 2-photon visual guidance in vivo. A short voltage pulse train is applied through the pipette to deliver RNA molecules into a targeted cell. The video below shows this process in cortical neurons expressing GCaMP6s.
