Apropos of nothing.... Peer-review should be rigorous, not rude. Critique the work, not the author team. Explain, don’t sneer. Build science up, don’t beat the authors down. Respect in peer-reviews isn’t optional — it’s how good research gets better.

We used synthetic DNA modifications to expand the alphabet of #nanopore sequencing. Write and read: Harnessing Synthetic DNA Modifications for Nanopore Sequencing @nanoporetech.com www.biorxiv.org/content/10.1... Here are five flavors of cytosine with characteristic current profiles
(A) The molecular conversion of cytosine into various natural and synthetic derivatives: 5-methylcytosine (5mC), 5-hydroxymethylcytosine (5hmC), 5-glucosyl hydroxymethyl cytosine (5gmC), and 5-glucosyl azide-hy-droxymethylcytosine (5gmC-N3). (B) Illustration of a single-strand DNA with modified cytosine passing through a nanopore. (C) A density plot showing the distribution of signal shifts of a single k-mer (GATGCG) demonstrating clear differentiation between modifications. Look at this crazy sarcoma genome! So many translocations, and it thrives! We were able to assemble it due to the ultra long optical genome mapping reads (N50>150kb). We mapped genomewide #DNAdamage before and after oxidative stress. Its not random! www.sciencedirect.com/science/arti...
