We conclude that TCR-focused computational approaches are becoming a central pillar of precision immuno-oncology, but realizing their full potential will require tighter integration of experimental and computational methods, larger datasets, and shared community standards.

AI is having a major impact on the field, particularly in structural modeling and specificity prediction. At the same time, many of the key bottlenecks remain biological: limited ground-truth datasets, TCR cross-reactivity, benchmarking, and generalization to unseen antigens.

One major challenge is that TCR analysis is no longer just a sequencing problem. Understanding T-cell responses increasingly requires integrating repertoire sequencing, structural biology, antigen specificity, and clinical data into a coherent framework.
