It’s finally starting to feel real 🧬🌾 Preprint's out! @daganlab.bsky.social we asked: when does a plasmid stop being “accessory”? In Pantoea agglomerans, LPP1 promotes wheat-root colonization and is deeply integrated with chromosomal gene regulation and metabolism. www.biorxiv.org/content/10.6...

This suggests that domesticated plasmids in Pantoea arose from large ancestral plasmids that experienced genome streamlining, similar to that observed in endosymbionts.

By using phylogenetics, we show that most genes with homologs in both the plasmid and the chromosome have different evolutionary origins. Moreover, we identify related plasmids outside of the genus that are rather large.

Devani Romero Picazo Interestingly, plasmid LPP-1 harbours genes that are core in the genus pangenome. Most of these genes encode for transcriptional regulators, suggesting the integration of the plasmid in the host's regulatory network.

These domesticated plasmids have diversified within the genus, where plasmid groups -with different gene content and sizes- coincide with main speciation events.

We show that the genomes of plant-associated Pantoea comprise two domesticated plasmids that are inherited vertically, just like chromosomes. We prove that the largest plasmid (LPP-1) replicates synchronously with the chromosome.
