In the E5.5 epiblast, WT1 expression and splice composition align with lineage-biased transcriptional states. This suggests lineage-associated programmes begin to diversify before gastrulation, with WT1 acting as an isoform-tuned regulator of early lineage bias. (3/4)

WT1 biases transcriptional programmes and is sufficient to drive post-implantation identity. Alternative splicing produces WT1 isoforms with distinct effects: → one biases anterior / neural programmes → another biases posterior / mesodermal programmes (2/4)

When do lineage biases first emerge in the epiblast? They are detectable before gastrulation, but how this early asymmetry is established has remained unclear. A CRISPRa screen identifies WT1 as an unexpectedly early regulator of formative pluripotency. (1/4)
